Description

uberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis and is still a global health threat to humanity. The World Health Organization estimates there were more than 10 million new cases and reported more than 1.5 million deaths in 2019, making TB one of the leading causes of death from a single pathogen. Standard anti-TB therapy includes four first-line antibiotics that must be given for at least six months. However, in cases of multidrug-resistant and highly drug-resistant TB, second-line drugs must be used and these drugs often cause severe side effects resulting in poor compliance.Therefore, developing new anti-TB drug candidates is of utmost importance. In this Special Issue dedicated to Tuberculosis Drug Discovery and Development, we present the main and recent achievements in the fields of drug and target discovery, host-directed therapy, anti-virulence drugs, and describe the development of two advanced compounds: macozinone and delpazolid. In addition, this Special Issue provides a historical perspective focusing on Carlo Forlanini, the inventor of the pneumothorax for the treatment of tuberculosis, and includes an overview of the most advanced technologies currently utilized in the development of tuberculosis drugs.
